Investment in Infrastructure
The HSC recently invested in the purchase of an AB Sciex QTRAP 5500 liquid chromatography tandem mass spectrometry system (QTRAP-LC/MS/MS) to build capacity in the Metabolomics Core Facility. This instrumentation complements the Thermo Scientific Trace 1310 TSQ8000 gas chromatography tandem mass spectrometry system (GC/MS/MS) capable of electron and chemical ionization, located in the facility and Thermo Fisher Q Exactive high-performance benchtop quadrupole-Orbitrap LC-MS/MS, which is located in the BioNano Research Facilities. The Mitochondrial Functional Assessment Core (MFAC) has the capacity to assess mitochondrial respiration and cellular glycolysis (Seahorse Bioscience XFe 96 instrument), mitochondrial membrane potential, activities of each of the complexes in the electron transport chain, fission-fusion dynamics, Ca2+ flux, reactive oxygen species production, and oxidative damage to lipids, proteins and DNA.
The HSC supports a home-cage Chronic Laboratory Animal Monitoring System (CLAMS), which allows measurement of whole body metabolism (via indirect calorimetry), food consumption, physical activity and temperature and heart rate by telemetry. This developing infrastructure provides the capability to correlate changes in the metabolome, alterations in glucose metabolism and mitochondrial respiration and whole body metabolism.
